Data on the frequency of these mutations in the pediatric population is limited. The aim of presented study was to determine the frequency of the USP8 gene mutations in a group of pediatric patients with CD treated at the Children's Memorial Health Institute (CMHI).
Eighteen patients (9 females) with CD were treated at CMHI, Warsaw, Poland between 1993 and 2019. All patients underwent transsphenoidal surgery (TSS) as a primary treatment for CD and were operated by the same neurosurgeon at CMHI. The average age of all patients at TSS was 13.10 yrs. (5.42-17.25). DNA was extracted from formalin-fixed paraffin embedded resected tumor tissue. Sanger sequencing was performed on DNA sequence corresponding to the exon 14 of USP8 gene.
The mean age at diagnosis of CD was 13.08 years and an average duration of symptoms before diagnosis 2.96 years. All patients were operated at CMHI by the same neurosurgeon. Fifteen out of eighteen patients (83.33%) had the initial biochemical remission after a single TSS procedure (post-operative serum cortisol &lt;1.8 μg/dl). The result of genetic testing was negative for all samples at the hotspot area of the USP8 gene.

Thyroid carcinoma RNA-Seq dataset from the TCGA database was used to assess the expression of SEMA6B in tissues, as well as its clinical significance. We adopted qRT-PCR and western blot analyses to measure the mRNA and protein expression of SEMA6B in thyroid carcinoma cells. The biological roles of SEMA6B in thyroid carcinoma cells were examined through cell counting kit 8, clone formation, and Transwell assays. Also, GSEA was used to identify the gene sets modulated by SEMA6B, which is further verified by western blot.
According to the public dataset from the TCGA database, we found that the expression of SEMA6B was upregulated in thyroid carcinoma tissues compared to adjacent non-tumour tissues, and a high level of SEMA6B resulted in a poorer prognosis compared to the low-level SEMA6B group. Functional experiments showed that silencing SEMA6B suppressed the B-CPAP cells viability, invasiveness, and motility, whereas up-regulating SEMA6B in FTC-133 cells led to opposite outcomes. Furthermore, knockdown of SEMA6B in B-CPAP cells could significantly elevate the protein expression of NUMB and reduce the expression of NOTCH1, HES1, and Cyclin D1. Conversely, overexpression of SEMA6B in FTC-133 cells presented opposite results on the protein expression of these Notch signalling pathway-related markers.

From January 16, 2020 to March 28, 2020, among the 568 cases of COVID-19 patients diagnosed in Xiaogan Central Hospital, 64 cases of COVID-19 patients with diabetes were selected as the diabetic group, and 64 cases of COVID-19 patients with age and gender matching without diabetes were selected as the non-diabetic group, and their clinical data and pulmonary CT characteristics were retrospectively analysed.
Compared with the non-diabetic group, the proportion of patients in the diabetic group with chronic underlying disease was higher, and they were in more a serious condition at admission. Inflammation index and characteristics of glycolipid metabolism results showed that COVID-19 patients with diabetes mellitus were more likely to have elevated inflammatory markers and hypercoagulability, accompanied by hypoproteinaemia and glucose and lipid metabolism disorders.
